Sequence Related Amplified Polymorphism (SRAP) is also known as Sequence Based Amplified Polymorphism (SBAP). This marker amplifies ORFs, which are important components of genes, through a pair of uniquely designed forward and reverse primers, and the genetic diversity of ORFs better reflects the diversity of genetic resources. Since the reverse primer used in SRAP markers contains AATT at the 3' terminal to specifically bind the AT-rich region. Such a unique primer design makes it possible to amplify intron and exon-based SRAP polymorphic markers. At the same time, the primers are longer, 17 or 18 bp, and can be used in combination, which greatly reduces the cost of primer development.

Therefore, SRAP can be used in many fields such as genetic diversity analysis, genetic map construction and gene localization, identification of good germplasm and varieties, and prediction of hybrid advantage due to the advantages of easy operation, good reproducibility, efficient co-display, and easy cloning and sequencing.
SRAP markers show relatively more informative bands both at the DNA level and at the cDNA level, thus providing a more comprehensive indication of the degree of polymorphism in the genome.
