Yeast monohybrid (Y1H) is a technique developed from yeast two-hybrid, mainly used to study the interactions between proteins and DNA elements. The transcriptional activator of many eukaryotes consists of a physically and functionally independent DNA binding region (BD) and a transcriptional activation region (AD). Thus fusion expression vectors for various genes with AD can be constructed. When expressed as a fusion protein in yeast, the protein with a specific binding region to the target element can then be screened based on the expression of the reported gene.
Lifeasible offers Y1H technology service for our customers to study the interactions between biological macromolecules. This service includes a complete experimental workflow covering library construction, one-hybrid screening, and one-to-one validation services.
We use pGADT7 and pGADT7-Rec2 systems for screening. Customers can use pre-made commercial libraries or our custom-built yeast hybrid libraries. We have many years of experience in library construction and screening and have completed thousands of libraries of various types, which can effectively ensure the coverage of libraries and the detection of target genes for library screening.
One-to-one validation service
In addition, we provide further validation solutions for Y1H results, including EMSA (probe synthesis and labeling and EMSA experiments) and dual fluorophore enzyme assay (vector construction and dual fluorophore enzyme assay) to further verify the interaction between gene promoter and protein.

You only need to provide information on the decoy plasmids used for screening, and Lifeasible will complete the entire screening process.
