Riboflavin is a member of the B vitamin group, which is an essential nutrient for mammals. In the last decade or so, the chemical synthesis of riboflavin has been completely replaced by microbial synthesis. In recent years, the production cost of riboflavin has continued to rise. Therefore, further improvement of riboflavin production strains and reduction of fermentation production costs have become urgent.
Lifeasible used synthetic biology techniques to construct an engineered strain of E. coli with high riboflavin production, to structurally optimize and chemically synthesize multiple genes of the ab initio biosynthetic pathway of riboflavin in Bacillus subtilis, and to reconstruct the riboflavin biosynthesis system in E. coli. Ultimately, in vitro targeted molecular evolution of the relevant RNA polymerase, riboflavin ab initio biosynthesis system, and global transcriptional regulators will be performed by DNA rearrangement technology to obtain an engineered strain of E. coli with excessive riboflavin production.
