Hordeum vulgare is an important food crop. Currently, the main means of Hordeum vulgare selection is still traditional breeding, which is time-consuming and requires a lot of human and material resources. With the rapid development of molecular biology technology, transgenic technology has been recognized as one of the effective techniques to innovate crop germplasm and select new varieties. Establishing and optimizing the Agrobacterium-mediated genetic transformation system for Hordeum vulgare provides a reliable solution for variety selection and promotion of its research and application. We used young and mature embryos as explants and compared the effects of different embryo treatments, different basal media, different carbon sources, and different hormone ratios on the induction and differentiation of healing wounds to screen and determine the suitable receptor materials for the genetic transformation of Hordeum vulgare, and to derive the optimal transformation conditions. In addition, we compared the effects of different genotypes on the healing and differentiation rates based on the optimized parameters to broaden the receptor genotypes of Hordeum vulgare genetic transformation and lay the foundation for establishing a further barley genetic transformation system.
Based on the establishment and optimization of the regeneration system of Hordeum vulgare mature embryo, we introduced the β-glucuronidase gene (i.e., GUS gene) into the healing tissues of Hordeum vulgare mature embryo by Agrobacterium-mediated method and optimized the Agrobacterium-mediated genetic transformation system of Hordeum vulgare through the parameter screening. The Hordeum vulgare genetic transformation system we established, using Hordeum vulgare seeds as transformation recipients, could successfully obtain transgenic Hordeum vulgare-positive materials and cultivate Hordeum vulgare transgenic plants by inoculation of prickly embryos with Agrobacterium rhizogenes.
